Primary antibody was tested in human, mouse and marsupials spread spermatocytes at different dilutions: 1/30, 1/100,1/200 and 1/300 (stock concentration: 0.77 mg/ml). Slides were incubated at 4?C and 37?C O.N. in humid chamber.
Secondary antibodies: goat anti-mouse IgG FITC and TRITC Conjugate (Sigma). Working dilution: 1/50 (stock concentration: 1mg/ml)

Thank you for your interesting feedback on ab6000. Recent publications have shown that sperm chromatin is highly specialised and the paternal genome is packed by small core-histone like proteins (minor variants of somatic histones) and protamines. This antibody specifically recognizes Histone H3 (di+tri methyl K4) but not the histone variants in spermatocytes.
