Additional Notes
The same chromatin was used for another ChIP assay using another antibody (ab6002), in parallel to the one of this trial, as a positive control.
The 2 independent experiments were performed in exactly the same conditions, using about 10*10e6 cells, 10ug of antibody and 80ul Dynabeads protein A (invitrogen 10002D).
The DNA recovery and enrichment of the control ChIP was good, while for the ab24175 ChIP the DNA recovery was undetectable by Qubit dsDNA HS assay kit (invitrogen, Q32854).
Therefore it was impossible to use any other detection method such as qPCR to validate ab24174 for ChIP.
Abcam response
Thank you so much for testing this antibody in ChIP on mouse samples.
We appreciate your effort and feedback, but are sorry to hear that the antibody did not work in this species/application. As stated on the datasheet, we do not recommend this antibody for use in ChIP or on mouse samples. We would welcome additional feedback from other customers as well.
The reviewer received a reward from Abcam’s Loyalty Program in thanks for submitting this Abreview and for helping the scientific community make better-informed decisions.
