thanks for your fast reply. The Abcam HRP antibody didn't give any signal (more than some black spots) but the same primary antibody worked nicely at the same time point with another secondary antibody. We tried to dissolve the antibody, but as you can see on the pictures there's still remaining significant clumps. Can you therefore, please send me the ab97051 as a replacement vial. Best regards, |

we were really unhappy to find out that the antibody weordered (date: 13thof April 2012) from Abcam (order number:1070469,lot number: GR27131)have precipitated, (se attached pictures).When we anywaytried to do thewestern blot unfortunately, this didn’t work.This is extra unfortunatebecause we had to usethe antibody for a demo of western blot imagingsystem. As you can se on the attached images the antibody forms a significant pellet (Pic_I) and when we tried to dissolve it it still large clumps in the solution (Pic_II). Thank you for your email. I am sorry to hear about the inconvenience caused. We are aware of this problem and doing our best to resolve this issue. We generally recommend our customers dissolving the lumps and using the antibody as normal, in many case we have even asked customers to centrifuge out the precipitates and for many customer the antibody has worked perfectly well. There are however inconsistencies with the complaints we receive from our customers. Although small lumps are very common with HRP conjugated however because this is a constant issue with this antibody so we are interested in collecting more feedback. Could you please provide some details about the problem; - Does this antibody gave high background, multiple bands or no staining? - Have you tried any sort of troubleshooting to optimize the experiment? - Is the primary antibody fine? Have you tried the primary antibody before or with different secondary antibody. Finally, would you like to try the antibody ab97051 or ab97069 as replacement. En principio el precipitado amarillo presente en el vial no debería aparecer, y los viales que habéis recibido lamentablemente si lo tenían.
Puedo mandaros dos nuevos viales confiando en que no vuelva a pasar (que es lo normal), o si preferís cambiar de anticuerpo ya que el anterior no os funcionó, puedo mandaros uno distinto de las mismas características, siempre y cuando me indiquéis las categorías que os pregunté en el mail anterior, que son necesarias para poder sugeriros la mejor alternativa.
Así pues, lo dejo a vuestra elección, y quedo a la espera de que me lo indiquéis para proceder con el pedido. |
