This antibody produced a membrane bound staining in cortical neurons. Nothing else was stained. The pictures show the staining obtained at the level of the cerebral cortex, at low power magnification (left panel) and higher power magnification (right panel). This staining is not consistent with previous observations from the human database (http://www.proteinatlas.org/tissue_profile.php?antibody_id=12465&g_no=ENSG00000163812) or from the allen institute (http://mouse.brain-map.org/brain/gene/71763655/thumbnails.html). There should not be any staining in cortical neurons, but a strong staining in the cerebellum.
The sections used came from animals perfused fixed with Paraformaldehyde 4%, in phosphate buffer 0.2M. Following postfixation in the same fixative overnight, the brains were cryoprotected in sucrose 30% overnight. Brains were then cut using a cryostat and the immunostainings were performed using the ‘free floating’ technique.
