Cryopreserved baboon ovary was thawed in M199 media and fixed in 4% paraformaldehyde, dehydrated in 70% ethanol, run through a tissue processor to permeate with wax, and embedded in paraffin wax. Blocks of tissue were sectioned 6´m thick and placed onto slides. Slides were dried, deparaffinized, brought through antigen retrieval, blocked and stained. Staining was with Rabbit anti GDF9 polyclonal antibody (Abcam #ab93892, lot# GR181977-1) at 1:200 followed by Goat anti Rabbit Alexa Fluor at 5´g/ml and Hoechst at 1´g/ml. Stained sections were mounted under coverslips using ProLong Gold solution. Once mounting media set, slides were imaged on an inverted fluorescence microscope.
