Neural Stem Cell Differentiation Protocol: Human ESCs and human iPSC line were maintained and differentiated according Stem Cell Technologies mTeSR's protocol and acclimated to the same culture condition for several passages before differentiation. After separation from feeder cells and culture in suspension for 7 days, aggregates of human iPSCs or hESCs were differentiated to primitive NEs in an adherent culture in the neural medium consisting of DMEM/F12, 2% N2 supplement, and 1% nonessential amino acid. Neural tube-like rosettes at day 15 of differentiation were then detached mechanically and cultured in suspension in the same medium. FGF2 (20ng/mL) and Noggin (5ng/mL) were added to cultures for the first 15 days, 10uM SB43152 was added from day 0–5.
