The image was produced using a sheep anti-rat secondary conjugated to biotin with streptavidin-Alexa 647 for fluorescent imaging. The signal is weak for small vessels, but using either an ABC peroxidase-based or biotin-streptavidin based amplification detection, it is possible to use this CD31 for capillary staining. We chose far-red fluorescent signaling due to the higher signal:autofluorescent noise in that spectrum.
