Peripheral Blood Monocuclear cells were isolated from fresh human blood and fixed in suspension for 20 min at room temperature (RT) in 2% paraformaldehyde solution. Cells were washed andblocked with 2% goat serum in pbs for 15 min. After centrifuging supernatant was aspirated and stained with CD14-FITC antibody for 45 min at RT. Cells were washed three times with PBS and stained for nuclear stain Draq 5 and plated in Concanavalin A treated 96 well plates and images were acquired using widefield microscope using FITC filter at 40X magnification
