Cells were fixed for 15 min and permeabilized for 4 min. No other fixative was tried. The green co-staining was obtained using a monoclonal anti-GM130, a Golgi-associated protein. A negative control was performed using non relevant IgGs. This antibody stains the nucleus.

An option is to use methanol fixation to see if localization differs.
The concentration is unknown as this antibody is tissue culture supernatant. Apologies for the confusion.
Thus, dilutions 1/250 and 1/1000 should be tried instead.
